Previous studies have shown that macrophages play an important role in regeneration, including lens regeneration in the Pleurodeles waltl. We wanted to explore the unique dynamics between inflammation and macrophages during the lens regeneration process.<br>Lens removal surgeries (lentectomies) were performed alongside intraocular injections of PBS as the control, and clodronate as the macrophage-depleting drug. Eyes were collected at 30 days post-lentectomy (dpl). Our results show that in the absence of macrophages, lens regeneration not only fails but adopts a scarring response in place of a regenerative response. Due to the general association between scarring and inflammation, we used bulk RNA-sequencing of the whole newt eye at various time points after injury to investigate inflammatory markers. There was a significant upregulation of pro and anti-inflammatory markers after lentectomy. In order to investigate the importance of inflammation in the lens regeneration process, we treated newts with anti-inflammatory drugs FK506 and Celastrol at -2, 0, 2, and 4 dpl. Eyes were collected for histology at 40 dpl. The anti-inflammatory drugs did not halt lens regeneration as lenses still formed in all groups. These results could potentially stem from the drugs not being at a high enough concentration, or the drug delivery method not being optimized. In the future, we plan to further optimize the protocol by potentially increasing the dose of the drugs and\/ or try intraperitoneal or intraocular injections to see if the same response is elicited through an alternate drug delivery system. Additionally, gene expression studies will be performed to analyze whole eye tissues treated with FK506 and celastrol after injury to see if the drugs have any effects on down-regulating inflammatory cytokines.<br>Author(s): Joey Trapp, Sophia Carmen Ratvasky, Gerogios, Anthony Sallese, J.
