Calcium ion concentrations are implicated in medical conditions like hypercalcemia, which can lead to bone and kidney disease. Intracellular ion levels can be sensed using fluorescence indicators. This means we can quantify the concentration of calcium using light emission from indicator molecules. I will focus on two approaches for analyzing spectrum shape, a ratiometric analysis and a phasor analysis, and the advantages and disadvantages of each. From my literature review, the main advantage of the ratiometric analysis is that the calibration equation is based on a chemical model.
