PTSD is a debilitating condition that significantly reduces the quality of life and alters an individual\u2019s ability to regulate fear and safety cues. Rodent models and laboratory fear conditioning are effective in simulating PTSD conditions and allow for analysis of how exposure to trauma changes the brain and its ability to process and respond to future threats. The paraventricular nucleus of the thalamus (PVT) is a midline brain structure that integrates threat- and arousal-related signals from cortical and subcortical inputs and relays this information to limbic circuits that mediate defensive responses. The present study investigated a role for the PVT in mediating the expression of safety learning. We presented an auditory safety cue that predicted the absence of footshock within a context that strongly predicts footshock. We infused half of the rats with the GABA-A receptor agonist, muscimol, directly into the PVT prior to the test of safety learning. Data are currently being analyzed. If the PVT is necessary for the expression of learned safety, inhibition of this structure will reduce safety (i.e., eliminate the decrease in freezing observed in the presence of the auditory safety cue) compared to rats that receive a control infusion. If there is a trending sex effect, future studies will be conducted to address the possible sex differences in PVT and the expression of safety learning.
