Despite these amazing strides, secondary cataracts occurs at a staggeringly high rate, with children being at the greatest risk of undergoing a harmful fibrotic response after the surgery and additional complications. Interestingly, organisms such as the newt are able to regenerate a fully functional lens without undergoing fibrosis when the lens is injured or removed. Our lab investigates the mechanisms behind lens regeneration within the newt. Whether or not macrophages potentially foster this fibrotic-free response has not been explored. Therefore, we have developed a range of molecular tools and techniques to study newt macrophages. Briefly, our results show that purified newt macrophages can be isolated from the peritoneal cavity and that the newt lung and spleen can be used as macrophage rich tissues for testing antibodies for immunohistochemistry and western blot analysis. CSF1R is a macrophage-specific protein commonly used to identify macrophages in mammals. However, while previous attempts to identify newt macrophages using traditional antibodies for human or mouse CSF1R had failed, we found an antibody specific to the conserved kinase domain that successfully recognized macrophages in newt tissue sections. With these new tools, we have begun screening strategies for inducing macrophage depletion in newts. This will ultimately allow us to determine how lens regeneration is altered when macrophages are not present and if a fibrotic response is observed instead. In the future, we can apply our work to humans who suffer from these debilitating diseases and provide treatments and therapies that enhance their lives.
